sendo por isso considerado o cancro primário deste orgão. O hepatocarcinoma é o
terceiro tipo de cancro que causa maior mortalidade a nível mundial. Actualmente, as
opções primárias de tratamento do HCC consistem na recessão cirúrgica do tumor, no
transplante de fígado e na quimioterapia, apresentando qualquer uma destas opções de
tratamento múltiplas limitações. Assim sendo, existe uma necessidade urgente de
desenvolvimento de novas estratégias antitumorais, direccionadas para o HCC, que
apresentem uma elevada eficiência.
A terapia génica tem contribuído significativamente para o desenvolvimento de
estratégias antitumorais promissoras. Contudo, a sua aplicação clínica ainda se encontra
bastante limitada, nomeadamente devido à reduzida eficiência de entrega de material
genético às células alvo. Assim, a aplicação da terapia génica está dependente, em
grande parte, do desenvolvimento de sistemas de entrega de material genético mais
eficientes. O HCC é caracterizado pela sobreexpressão do receptor das
asialoglicoproteínas (ASGP), que é expresso exclusivamente no fígado. A asialofetuina
é um dos ligandos reconhecidos especificamente por este receptor. Deste modo, a
utilização deste ligando em sistemas de transporte de material genético poderá permitir
uma entrega eficiente e direccionada dos ácidos nucleicos no HCC.
O principal objectivo deste trabalho consistiu em desenvolver um novo sistema de
transporte e entrega de material genético, específico para células de hepatocarcinoma,
baseado em lipossomas catiónicos, compostos por EPOPC (1-palmitoil-2-oleoil-snglicero-
3-etilfosfocolina) e Colesterol, e em asialofetuina. Os resultados obtidos, nos
estudos de transfecção realizados na linha celular de hepatocarcinoma HepG2,
mostraram que a associação de asialofetuina aos lipoplexos promove uma potenciação
significativa da sua actividade biológica, nomeadamente para a formulação composta
por EPOPC:Colesterol/ADN, na razão de carga (+/-) 2/1, contendo 10μg de
asialofetuina/μg de ADN e para a formulação preparada com EPOPC:Colesterol/ADN,
na razão de carga (+/-) 4/1, contendo 20μg de asialofetuina/μg de ADN. Este aumento
substancial da actividade biológica dos lipoplexos, induzido pela associação de
asialofetuina, foi verificado não apenas pelo aumento da expressão do transgene
(quantificação da expressão da luciferase) mas também pelo aumento da percentagem de células transfectadas (quantificação das células que expressavam a proteína verde
fluorescente). Os resultados obtidos mostraram também que a actividade biológica
registada com os complexos preparados com asialofetuina, em células HepG2, foi
superior à verificada com formulações que apresentaram, em diversos estudos, elevada
actividade biológica em vários tipos de linhas celulares. O ensaio de protecção de
material genético mostrou que a presença de asialofetuina nos lipoplexos resulta num
menor acesso do brometo de etídeo ao ADN dos complexos, constituindo um indício da
sua maior protecção. Na presença de galactose, que compete com a asialofetuína pela
ligação ao receptor ASGP, verificou-se uma diminuição significativa da actividade
biológica dos lipoplexos preparados com asialofetuina, não tendo sido, no entanto,
observada uma redução da capacidade de transfecção para as outras formulações de
complexos. Os resultados registados nos estudos de ligação e associação celular
demonstraram que a presença de asialofetuina induz um aumento da ligação e da
internalização dos lipoplexos nas células HepG2, sendo esse aumento parcialmente
inibido pela presença de galactose. Os resultados obtidos neste trabalho demonstram
que estes lipoplexos, preparados com lipossomas catiónicos de EPOPC:Colesterol e
asialofetuina, constituem um novo sistema de transporte e entrega de material genético
que poderá ser de grande importância para o desenvolvimento de novas estratégias
antitumorais contra o HCC. Hepatocellular carcinoma (HCC) is originated in the main liver cells, the hepatocytes, being considered the primary cancer of this organ. Hepatocellular carcinoma is the third largest cause of cancer mortality worldwide. Currently, the primary HCC treatment options consist in tumor surgical recession, liver transplantation and chemotherapy. However, all these treatment options present multiple limitations. Therefore, there is an urgent need to develop new antitumor strategies, targeting HCC and presenting high efficiency. In the last years, gene therapy has significantly contributed to the development of promising antitumor strategies. However, its clinical application is still limited, mainly due to the reduced efficiency of genetic material delivery into target cells. Therefore, the application of gene therapy is largely dependent on the development of efficient gene delivery systems. On the other hand, HCC is characterized by the overexpression of the asialoglycoprotein receptor (ASGP-R), which is exclusively expressed in the liver. Asialofetuin is a ligand that specifically binds to this receptor. Regarding this, the use of this ligand in gene delivery systems may allow an efficient and targeted release of nucleic acids in the HCC. The main goal of this work consisted in the development of a new gene delivery system, specific for hepatocellular carcinoma cells, based on cationic liposomes, composed by 1-palmitoyl-2-oleoyl-sn-glycero-3-ethylphosphocholine (EPOPC) and cholesterol, and asialofetuin. The results obtained in transfection studies, performed in the hepatocellular carcinoma cell line HepG2, showed that the association of asialofetuin to lipoplexes promotes a significant increase in their biological activity, this being particularly evident for the lipoplex formulations EPOPC:Cholesterol/DNA, at the 2/1 (+/-) charge ratio, containing 10μg of asialofetuina/μg DNA, and EPOPC:Cholesterol/DNA, at the 4/1 (+/-) charge ratio, containing 20μg of asialofetuina/μg DNA. This substantial enhancement in the biological activity of the lipoplexes, induced by the association of asialofetuin, was observed not only by the increase of the transgene expression (luciferase expression quantification) but also through the enhancement of the percentage of transfected cells (quantification of cells expressing the green fluorescent protein). The results also showed that, in HepG2 cells, the biological activity of these complexes prepared with asialofetuin was higher than that obtained with other formulations known for presenting high biological activity in different cell lines. Regarding the assay on the protection of genetic material, it was observed that the association of asialofetuin to lipoplexes results in a lower access of ethidium bromide to DNA of the complexes, providing an evidence of its higher protection. In the presence of galactose, which competes with the asialofetuin for the binding to the ASGP receptor, it was observed a significant decrease in the biological activity of the lipoplexes containing asialofetuin. This reduction was not observed for the other complexes formulations. The results obtained in the binding and cell association studies showed that the presence of asialofetuina induces an increase in the binding and internalisation of the lipoplexes in HepG2 cells, this increase being partially inhibited by the presence of galactose. The results obtained in this study show that these lipolexes, prepared with EPOPC:Cholesterol cationic liposomes and asialofetuin, constitute a new gene delivery system that may be of great importance for the development of novel antitumor strategies against HCC. |
